Memasuki paruh musim MotoGP 2020, sejumlah tim sudah bersiap menyambut kompetisi tahun depan dengan mengubah susunan pebalap mereka. Sejumlah perubahan line up pebalap untuk MotoGP 2021 sudah terjadi. Ducati dan Yamaha menjadi tim yang melakukan perubahan terhadap line up pembalap mereka.
Ducati melakukan switch pada Pramac Racing dan Monster Energy Yamaha juga melakukan switch pada tim Petronas SRT Yamaha. Di musim 2021 Ducati Corse selaku tim pabrikan bakal diperkuat oleh Pecco Bagnaia dan Jack Miller menggantikan Andrea Dovizioso dan Danilo Petrucci. Sedangkan Rossi bakal bertukar ke Petronas SRT Yamaha menggantikan Fabio Quartararo yang naik ke tim pabrikan Yamaha, Monster Engery Yamaha.
